Is it able to add another instant without using the CD to setup again?

Is there any way to add the instant in the Enterprise Manager?

Yes and No...(you're actually asking 2 different questions...:-)...).
If you want to install an additionally Named Instance on your SQL
server, you need the CD. A new instance is more or less to be seen as a
new installation.
If you just want to register an already installed Instance in EM, you
can do it without the CD. Here you just do a new SQL Server registration .
